Highlights
•Fatty acylation of free remdesivir (RDV) offered two structural conjugates.
•2D H–HCOSY, HSQC, and HMBC confirmed the structure of each isomer.
•Conjugates of RDV with fatty acids were evaluated against SARS-CoV-2 and EBOV.
•3′-(4-Oxatetradecanolyl) of RDV was most effective against SARS-CoV-2.
•Some fatty acyl conjugates of RDV showed comparable anti-EBOV activity with RDV.
